Donnie Murphy was first drafted in 2002 by the Kansas City Royals. He played in the Royals' minor league system from 2002 through 2006, playing with the Royals in just 7 games in 2004 and 32 games in 2005 before returning to the minors for the entire 2006 season.

The Oakland Athletics acquired Murphy in November, 2006. He continued spending most of his time in the minor leagues, playing in 42 games with the Athletics in 2007 and 46 games in 2008. In 2009, he signed a minor league contract with the Baltimore Orioles and he played in their minor league system that year.

In December, 2009, Murphy signed a minor league contract with the Miami Marlins. Unfortunately for both the Marlins and Murphy, he suffered a wrist injury and had surgery in 2010. His playing time with the Marlins was limited to 29 games in 2010 and 36 games in 2011. The 2012 season was not much better, with Murphy again spending most of his time on the DL and in the minors, playing in just 52 games with the Marlins.

Murphy signed a minor league contract with the Milwaukee Brewers in December, 2012, but he was released in March, 2013. A month later, he signed with the Chicago Cubs. Last year, he batted .255 in 46 games with the Cubs. After the 2013 season ended, Murphy signed a one-year contract with the Cubs. In March, 2014, he was acquired by the Rangers.

After 45 games, the Rangers released Murphy on July 9, 2014. He had 22 hits and a .196 batting average in his time with the Rangers.

Murphy signed with the Cincinnati Reds on July 18, 2014. Less than a month later, he was released by the Reds. On August 16, 2014, he was signed by the Atlanta Braves. He didn't play with either the Reds or the Braves in 2014.

Murphy became a free agent on November 3, 2014. He signed a minor league contract with the Milwaukee Brewers on February 18, 2015. After time on the DL and 55 games in the minors, he was released on August 2, 2015. He hasn't played professional baseball since 2015.

Murphy was a minor league coach for the Toronto Blue Jays in 2018 and 2019. He was made a minor league manager on January 31, 2020.
